Thymus development is critical for adaptive immune systems. Researchers such as Erik Kernfeld of the University of Massachusetts Medical School used single-cell RNA sequencing technology to observe embryonic thymocytes and T cell receptor rearrangement disorders that developed to 8 days. Studies have confirmed the identity of T cells in both conventional and unconventional lymphocytes, described the maturation kinetics of thymic epithelial cells in vivo, and revealed the intrinsic ability of thymic epithelial cells to maintain transcriptional regulation. By combining cell maps with human genome-wide association studies (GWAS) data and autoimmune disease-associated genes, they found that embryonic thymocytes may be participants in the cause of autoimmune diseases.
