The study found a new molecular mechanism of obesity associated with colon cancer

Obesity has become a global health problem, and there is ample evidence that obese people are at higher risk of colon cancer, and colon tissue inflammation is a major risk factor for colon cancer. A study published in the Proceedings of the National Academy of Sciences on the 30th revealed a new molecular mechanism by which obesity increases inflammation in colon tissue.

Zhang Guodong Laboratory of the University of Massachusetts Amherst and Bruce Hammock Laboratory of the University of California, Davis, found a "soluble epoxide hydrolase" (sEH) and inflammation of the colon tissue caused by obesity through mouse experiments. Relatedly, inhibition of this enzyme can control inflammation of the colon tissue, thereby reducing the risk of colon cancer.

The researchers used a metabolomics method based on liquid chromatography-mass spectrometry to find that sEH produced higher levels of fatty acid metabolites in the colon tissue of obese mice. Zhang Guodong, author of the paper, said: "Based on metabolomics data, we further found that the expression of sEH in colon tissue of obese mice was significantly increased."

The researchers used two different enzyme inhibitors and mouse validation that did not produce this enzyme after knockout, confirming that sEH plays a key role in the inflammation of the colon caused by obesity.

"After sEH, even in obese mice fed a high-fat diet, colonic tissue inflammation was blocked, indicating that sEH inhibitors may be an effective means of preventing colon cancer and rectal cancer caused by obesity." Doctoral student Zhang Jianan said.

Currently, sEH inhibitors have entered human clinical trials for the treatment of other inflammatory diseases. Phase I human clinical trials have found that these drugs are safe and have no side effects. Zhang Guodong said that these clinical resources will promote the future use of these drugs to prevent the risk of colon and rectal cancer caused by obesity, without the need to develop drugs from scratch.
